DETAILED PROGRAM
DAY 1
− Determination of assimilation/physiology profiles (assimilation of the main carbon and nitrogen sources) of yeasts, using 96 wells plate reader and growth rate calculation.
− Observation of macro-morphology (colour, form, elevation, margin, etc.) and micro-morphology (shape, size, a/sexual reproduction, pseudo/mycelia, presence of spores, etc.) of different yeast species.
DAY 2
Screening of technological properties of yeasts:
− production of extracellular enzymes (amylase, proteinase, pectinase, esterase): media preparation, inoculum and results reading and interpretation;
− resistance to antimicrobial compounds (calculation of the Minimum Inhibitory Concentration for ethanol): media preparation, inoculum and results reading and interpretation;
− definition of optimal temperatures in solid medium: set of the inoculum and results reading and interpretation.
DAY 3
Molecular identification of yeasts strains:
− DNA extraction using classical procedure and commercial kit;
− amplification of target region by PCR.
DAY 4
Molecular identification of yeasts strains:
− analysis of the sequences using Chromas Technelysium Pty Ltd;
− comparison with the Type strains sequences present in the free online database Blast (Basic Local Alignment Search Tool – http://blast.ncbi.nlm.nih.gov/Blast).
